Analysis of human cerebral organoids developed from both WA09 human embryonic stem cells with CRISPR-KO of NGLY1. The hypothesis tested was that NGLY1 is key to normal human brain development and when knocked out would alter cerebral development of organoids through the differential expression of various key genes. Overall design: Total RNA obtained from human cerebral organoids developed using WNT & TGFbeta inhibition (3uM IWR-1e & 10uM SB431542). Organoid samples were collected along the development timeline from the pluripotent stem cell stage to day 80 of cerebral maturation
