Spatial transcriptomic profiling of two replicates of Patient and two replicates of Patient-corrected bioengineered vascular grafts harvested at week 8 post-implantation. Overall design: A Loeys-Dietz syndrome-associated pathogenic variant in Patient induced pluripotent stem cells (PatientA230T/+) was corrected using CRISPR/Cas9 gene editing resulting in Patient-corrected (Patient+/+) cells. Patient-corrected (Patient+/+) samples are denoted as iWT in the sample identifiers. The resulting human induced pluripotent stem cells were differentiated into smooth muscle cells through cardiovascular progenitor cells. The smooth muscle cells were seeded on meshes to generate bioengineered vascular grafts. The bioengineered vascular grafts were implanted into nude rats and were harvested at week 8 post-implantation. Formalin-Fixed Paraffin-Embedded (FFPE) sections of the harvested vascular grafts were processed using 10X Visium Spatial Gene expression for FFPE. Human transcriptome probe kit was used for the gene expression measurements.
