Current Adaptive Immune Receptor Repertoire sequencing (AIRR-seq) methods resolve antibody (IG) transcripts with limited resolution of the antibody constant region. Here we implemented a novel near full length AIRR-seq method (FLAIRR-seq) that utilizes targeted amplification by 5' rapid amplification of cDNA ends combined with single molecule real time smart sequencing in order to generate highly accurate IG heavy chain repertoires from human donor peripheral blood mononuclear cells. FLAIRR-seq allows for characterization of IGHV, IGHD, IGHJ and IGHC region genes and alleles from expressed antibody repertoires. Data also includes a FLAIRR-seq time course of one individual hospitalized with severe COVID-19. Iso-Seq and FLAIRR-seq was performed with healthy donor isolated B cells.
