Project: PRJNA940073
The thermotolerant TTY23, acid tolerant AT22, and thermo-acid tolerant TAT12 yeast strains, generated by adaptive evolution experiments were grown at 30°C, pH 5.0 in the absence of acetic acid (optimal ancestral conditions). The mRNA of each strain was isolated and changes in expression were compared against the parental strain S288C. Overall design: The microarray YOMWG_15, constructed for the yeast strain S288C, following the fabrication of GPL18099, but using oligo set for S. cerevisiae and the protocols described here, was used in these experiments. This was elaborated by the DNA Microarrays Unit of the Institute of Cellular Physiology, in the National Autonomous University of Mexico The mRNA of each strain was isolated in the middle exponential phase of growth. Changes in gene expression were measured with microarrays using the SWAP method (Gomez-Mayen AP, Corral-Guille G, Riego‐Ruiz L (2019) Package ‘genArise’, retrieved from http:// bioco nduct or. org/ packa ges/ relea se/ bioc/html/genArise.html) DNA microarrays unit of the National Laboratory of Technological Support to Genomic Sciences (contributor).
