The authors have assigned confidence based on reporter gene activation. The Y2H interactions that can be identified by activation of three reporter genes HIS3, URA3 and lacZ were regarded as of higher confidence and classified as LacZ4 set. The Y2H interactions identified only with HIS3 and URA3 reporter genes were classified as lower confidence data set SD4.

9665 Clones from human fetal brain library and 2136 full length human ORFs were used for automated Y2H matrix interaction mating. 4,456 baits were checked against 5632 preys after cloning and discounting the autoactivators. The interactions were performed in a pooled array. A second interaction mating was performed to give 3269 confirmed interactions between 1064 baits and 1075 preys.
1) The protein ID's were obtained by parsing the gene name and description line of UniProtKB entries for the symbol and the accession number recovered. 2) The identifiers given were matched for UniProtKB entries using http://www.pir.uniprot.org/search/batch.shtml against UniProtKB and UniParc and the UniProtKB and UniParc accession numbers recovered. 3) Some of the sequences could be attributed only to mouse. The sequences with the mouse identifiers were matched for human proteins by sequence alignment. Interactions between all identified proteins are reported. |
