Family I1 is frequently referred to as the Kazal family of inhibitors, in reference to the pancreatic secretory trypsin inhibitor (now known as SPINK1, I01.011) that was discovered by Kazal et al. (1948). As a result of the work on the inhibitor units of ovomucoid by Michael Laskowski, Jr. and his colleagues, the mode of interaction of inhibitors in family I1 with peptidases has become the most thoroughly characterised of all such interactions (e.g. Laskowski et al., 2003; see also Laskowski mechanism).
Peptidases inhibitedPeptidases inhibited belong to families S1 and S8 (Laskowski & Kato, 1980; Lu et al., 1997). They include endopeptidases with a broad range of specificities.
Mechanism of inhibitionInhibitors in family I1 inhibit serine peptidases by the Laskowski mechanism also termed the 'standard' mechanism (Laskowski & Kato, 1980).
Molecular structureThe family contains compound inhibitors; numbers of inhibitor units include 1 (e.g. I01.018), 2 (e.g. LI01-003, LI01-007), 3 (e.g. LI01-001), 4 (e.g. LI01-006), 5, 7 (e.g. LI01-002), 9 and 15 (e.g. LI01-004) (Rawlings et al., 2004).

Biological functionsSPINK1 (I01.011), the pancreatic secretory inhibitor, is packaged in the pancreatic granules, and is secreted with trypsinogen and other endopeptidase proenzymes in the pancreatic juice. It is thought to prevent premature activation of the peptidase proenzymes in the pancreas itself (Chen et al., 1996).
