Parasitic arthropods that feed on mammalian blood prevent coagulation to prolong feeding. The tick Haemaphysalis longicornis produces inhibitors in its saliva known as madanins which inhibits thrombin (S01.217) (Iwanaga et al., 2003).

Mechanism of inhibitionMadanins prevent conversion of fibrinogen to fibrin, the activation of coagulation factors V and VIII and aggregation of platelets, but not the amidolytic action of thrombin. Therefore madanins bind to the thrombin exosite 1 and not to its active site (Iwanaga et al., 2003).
