The first peptidase in the family was discovered in a virus that affects the Black Tiger Prawn (Penaeus monodon) (Cowley et al., 2000).

Active siteResidues His2879 and Cys2968 (see the Alignment) were identified by mutagenesis and suggested to form a simple catalytic dyad; no evidence was obtained for a catalytic triad involving an Asp residue (Ziebuhr et al., 2003).
Activities and specificitiesA tentative cleavage consensus site has been defined as -Val-Xaa-His-GluYaa, in which Yaa is Leu or Val (Ziebuhr et al., 2003). The specificity is quite distinctive, but the P2 His specificity is seen also in some plant potyvirus NIa peptidases (family C4).
Molecular structureIn sequence, the peptidase is closest to the viral 'main' endopeptidases of coronaviruses (family C30), but the specificity is more like that of enzymes from plant potyviruses.
