Only hyicolysin (M30.001), the peptidase from Staphylococcus hyicus, has been characterized (for a review see Götz, 2004).

Active siteHyicolysin is a metallopeptidase with a single zinc ion, liganded by two histidines and a glutamate. The histidines occur in an HEXXH motif, in which the glutamate is believed to be a catalytic residue.
Activities and specificitiesHyicolysin is active between pH 7.4 and 8.5. The peptidase is able to cleave azocasein and gelatin, but not elastin, FA-Gly-Leu-amide, a synthetic substrate for thermolysin (M04.001) nor FA-Leu-Gly-Pro-Ala, a synthetic substrate for collagenase 1 (M10.001).
InhibitorsHyicolysin is inhibited by the metal chelators EDTA, EGTA and 1,10-phenanthroline. Only zinc and cobalt ions can reactivate the enzyme. Unlike thermolysin, hyicolysin is not inhibited by phosphoramidon.
Molecular structureNo tertiary structure has been determined for any member of the family, but on the basis of the presence of the HEXXH zinc-binding motif, coupled with Glu as the third zinc ligand, family M30 is included in clan MA.

Biological functionsPhysiological substrates are unknown. Hyicolysin is synthesized as a precursor with an N-terminal signal peptide followed by a propeptide. Maturation is assumed to be by autolysis, and mature hyicolysin is secreted to the extracellular medium.
Pharmaceutical and biotech relevanceStaphylococcus hyicus infects pigs, cattle and chickens. In pigs it is associated with exudative dermatitis, necrosis of the tips of the ears, and tail-biting. Because the hyicolysin gene is present in isolates from infected animals, it is assumed to have a role in pathology and to be of agricultural and veterinary importance.
